Characterization of sperm motility in sea bass: the effect of heavy metals and physicochemical variables on sperm motility ArchiMer

Computer assisted sperm analysis (CASA) was used to characterize the motility of sea bass Dicentrarchus labrax spermatozoa and to study the effect of several physicochemical variables and heavy metals on sperm swimming performance. Duration of sperm motility in sea bass was very short (< 50 s). During the first 20 s all the motility variables measured remained approximately constant, the velocity and linearity of the movement being maximum during this period, while both variables decreased sharply later. While slight variations in pH did not significantly modify sperm swimming performance, changes in osmolality affected all the measured motility variables. Movement activity and localization of food were studied dynamically with the objective of establishing the capacities of behavioral adaptation of the young Dicentrarchus labrax faced with a novel situation. The role played by previous rearing conditions was demonstrated, the level of stimulation being a sensitive intervening factor (currents associated with the food). The data show the favourable effects of such treatment, and are in agreement with work carried out on higher vertebrates, which have brought to light the importance of early stimulation in behavioral ontogeny which respect to emotionality as well as capacities of adaptation in subjects reaching adulthood.

In the three species Dicentrarchus labrax, Sparus auratus , and Anguilla anguilla an artificial feeding (dry pellets) disturbs liver cells as revealed at ultrastructural level. In hepatocytes, anomalies consist of lipids storage (liposomes) considered as deviation in fatty acids metabolism. Moreover are observed cellular alterations (mitochondrial vacuolization, nuclear pycnosis) related to degenerative process. It is of great interest to understand wether liver modifications are just a reaction to an unbalanced feeding or a true trophic pathology.

Estuaries and coastal areas are often subjected to much pollution, including pesticides and polycyclic aromatic hydrocarbons (PAHs) produced by anthropic activities. Bay of Marennes-Oléron is an estuarine coastal zone in which there is a high level of aquacultural production and this has adverse effects in aquatic organisms. In fact, PAHs, which are widely spread throughout aquatic ecosystems, have a low polarity and so there is a clear tendency to bioaccumulate in marine organisms (1). Impact of aquaculture on the genetic structure of Mediterranean populations of Dicentrarchus ArchiMer

A phylogeographic analysis based on 6 polymorphic microsatellites was performed on 15 sea bass samples. five from Western Mediterranean, seven from Eastern Mediterranean and three coming from French aquaculture stocks. Among the eastern samples, three did not cluster according to their geographic origin but rather with the occidental group. Furthermore. a somewhat lower allelic diversity was observed within these particular samples, indicating that they originated from a limited number of progenitors of foreign origin. What is the heritable component of spinal deformities in the European sea bass (Dicentrarchus labrax)? ArchiMer

Malformations in farmed fish are a prime research topic as it has a major impact on fish welfare and on the economical value of the product. In this study, the genetic component of spine deformities was estimated in European sea bass, a major species of Mediterranean aquaculture, with the use of a posteriori reconstruction of pedigree with microsatellites. Our population exhibited a very high malformation rate (81%),probably due to intense swimming of fish during pre-growing The heritability of global spine deformities was evaluated to be 0.21 +/- 0.04 on the underlying scale (033 +/- 0.06 for lordosis, 0.13 +/- 0.04 for scoliosis). Mature eggs were expressed from captured female and fertilised artificially with slightly bloody sperm, in dry conditions or in a humid atmosphere, both at 13 degree C. The best incubation conditions proved to be 13 degree C, with streptomycin and penicillin. Another sample was kept at 18 degree C. Hatching occurred after 4 or 5 days, larvae showing positive phototropism. The system of renewal of water and artificial food is described. The embryonic reserve drop of lipid disappeared between day 14 and 16. Etude morphologique du loup Dicentrarchus labrax L. de la region de Sete ArchiMer

Sampling of a population of D. Labrax was one with spear diving catches. Morphometric and meristic data were used to calculate a biometric index. This index showed that all the bass of the Sete area belong to Dicentrarchus labrax, even if they were black spotted on the upper part of the body. Morphology was slightly different according to sex, but sex determination was not always possible using morphometric data. This inshore population shows some morphological characteristics (branchiospine and lateral line scale number) different from the data collected on the same sp along the Maroc Atlantic coast by Gravier, 1961.

Symptoms recalling a nutritional pathology and more precisely a process of lipid peroxydation were observed during the last years in seabass Dicentrarchus labrax and other fishes cultured in tropical marine condition, Ocyurus chrysurus and Lutjanus analis . They included dark coloration, skin ulceration, lethargy, anorexy, emaciation. Histological examinations showed hepatic fatty degenerative lesions, pancreatitis, muscular degeneration and retinal atrophy. Additional vitamins E and C in the food suppressed that pathological symptoms. Sea bass Dicentrarchus labrax larvae were hatched and reared for 34 days in the laboratory to test a recent biochemical index (DNA/C) of larval fish nutritional condition. Variations in DNA/C ratio values were monitored during the whole larval ontogenesis of fed individuals and the effect of starvation was assessed at three developmental stages. Three main periods in the relative DNA content variations were determined in close connection with growth patterns during larval ontogenesis.

Induction of ovulation and stimulation of spermation by LHRH or an LHRH analog associated or not with pimozide. Effects on some teleost fish. ArchiMer

Effectiveness of LHRH and one of its analogs (LHRH-A = D-Ala super(6) ethylamide) on induction of ovulation and stimulation of spermiation is reviewed here. LHRH was also combined with pimozide, an anti-dopaminergic agent, to try to cancel dopamine inhibition of gonatotropin (GtH) release. LHRH-A alone (50 mu g/kg) did not initiate ovulation but only oocyte maturation. When pimozide (10 mg/kg) and LHRH-A (50 mu g/kg) were combined, gonadotropic hormone secretion was significantly increased, causing ovulation and stimulating spermiation. This combination advanced ovulation in trout. Trout egg quality was markedly altered by the pimozide treatment; it remains to be tested in carp. The ClC-3 chloride channel and osmoregulation in the European Sea Bass, Dicentrarchus labrax ArchiMer

Dicentrarchus labrax migrates between sea (SW), brackish and fresh water (FW) where chloride concentrations and requirements for chloride handling change: in FW, fish absorb chloride and restrict renal losses; in SW, they excrete chloride. In this study, the expression and localization of ClC-3 and Na+/K+-ATPase (NKA) were studied in fish adapted to SW, or exposed to FW from 10 min to 30 days. In gills, NKA-alpha 1 subunit expression transiently increased from 10 min and reached a stabilized intermediate expression level after 24 h in FW. Regulation of feed intake, growth, nutrient and energy utilisation in European sea bass (Dicentrarchus labrax) fed high fat diets ArchiMer

Three practical isoproteic (54% protein) diets were formulated to contain graded levels of crude fat (diet L: 10%, diet M: 20% and diet H: 30%). Each diet was assigned unrestrictedly to three and restrictedly to two replicate groups of fish (IBW 243 g). In unrestricted groups, increasing the dietary lipid level led to a significant decrease in voluntary feed intake without affecting growth rate. Problems related to the lack of functional swimbladder in intensive rearing of the seabass Dicentrarchus labrax and the sea bream Sparus auratus . ArchiMer

The primordial inflation of swimbladder normally occurs at a size of 4 to 5 mm in sea bream Sparus auratus and 5 to 6 mm in sea bass Dicentrarchus labrax . However, in intensive larval rearing conditions, lack of inflation is often observed and leads to the production of up to 80% of postlarvae whose swimbladder is not functional. They are, on the average, 20-30% smaller in weight than normal fry. Their survival rate can also be reduced by any kind of stress such as weaning, handling or hypoxic conditions. It is suggested that the selective mortality which affects the abnormal fish results both from a decrease in predatory efficiency and an increase in energetic needs.
